Abstract or Summary

This project looked at use of the comet assay method and HPLC to determine the detoxification of PhIP by gut microbiota. While no definitive results were obtained there was a great deal of optimisation of the method carried out. There was comet formation in both the slide comet assays and the high throughput comet assays carried out, which suggests with further development of the assays may be an efficient method of analysing DNA strand breaks. The PhIP was able to be identified using the HPLC method however, due to the low retention time of PhIP in the analysis any quantitative results would be skewed. With a slightly more method development the HPLC method could provide quantitative data on the detoxification of PhIP.
